The big idea

The sperm of older rats is different from that of younger rats in ways that may affect embryo development, according to recently published research.

Instructions that determine which genes will be turned on or off in offspring can be found in sperm. Epigenetic information is what these directions are known for.

The activity of many genes in sperm was affected by the age of the rats we studied. We found that age affects two epigenetic mechanisms, one of which is the altering of DNA to block specific genes.

Why it matters

People are having their first child later in developed countries. Over the past three decades, the number of men between the ages of 35 and 44 who are fathering children has increased. Children conceived by older fathers have a higher risk of health problems.

There are many different forms of cancer, as well as other disorders such as attention deficit/hyperactivity disorder.

The quality of information transferred by sperm decreases with age. We don't know if these changes occur through an accumulation of genes or something else. Epigenetic changes are thought to be a culprit.

How we do our work

We collected sperm from old rats in our experiments. The ages are between 20 to 25 and 40 to 45. The sperm of older rats had different epigenetic profiles than younger rats, and we analyzed it using methods based on DNA sequencing.

In older rats, 5,319 DNA sites had different levels of methylation. 5% of all rat genes may be affected by the number of sites we found.

Older rat sperm had different concentrations of smallRNAs, which can interfere with the expression of genes. 3.6% of the smallRNAs we identified were different in older rats. Changes in smallRNA can affect multiple genes.

The genes that were affected by these epigenetic mechanisms were identified using a computer program. The brain and other organs are regulated by most of the genes we identified.

The study only analyzed sperm, not offspring. Knowing which genes are likely to be turned off allows us to predict what will happen in the development of rat pups. If the same mechanism exists in humans, it may be possible for children conceived by older fathers to have a higher risk of certain health conditions.

What still isn’t known

Similar age related changes occur in human sperm. The study of human sperm is still in its infancy. Epigenetic information is flexible and can be altered by many lifestyle and environmental factors. We hope our research will help to develop future therapies that can address the health of children before they are conceived.
