The new trilogy will leave the Homecoming storylines behind and move on to new territory. The fans are referring to this as the college trilogy, where Peter and company are in college instead of high school, and that requires some new cast members.
I think he would get his own series of films outside of Holland's if he were to be introduced within a Spider-Man movie. In terms of a co-star that could run the entire trilogy? There is only one character that comes to mind.

Gwen Stacy seems impossible to avoid in this era, as such, fans are already starting to fantasy cast who they want to see in the role, as it seems unlikely that, multiverse developments aside, it's going to continue to be an undead Emma Stone from the Andrew Garfield films.
She would become a hero in her own right, as a result of the new Spider-Man films turning her into Spider-Gwen by the end.
I agree with at least one name being passed around by fans, which I believe is that of Anya Taylor-Joy, who I believe will be pursued for the role, given her surge in prominence after The Queen's Gambit, and her classic look that seems perfect for the character. We don't know if that would work out or not, but we will see.

We wouldn't be expecting an official casting announcement for the next trilogy until after Gwen Stacy arrives. We will see if Marvel sticks with some of the more obvious picks, or if they go in a different direction for Gwen.
