Bryan Gil, Spain's winger, was one of nine internationals on the Spurs side.
Tottenham was punished for not sending their star players home. Vitesse Arnhem, a Dutch side, won the Europa Conference League.
Maximilian Wittek, Vitesse's left-sided midfielder, converted a volley at the area's edge with a crisp conversion. This defeats the visitors' chances of moving on comfortably in Group G.
Spurs had Son Heung-min and Harry Kane, two key players, resting. They therefore fielded a different starting team to the one that beat Newcastle in Sunday's Premier League. This team lacked rhythm and put in a poor performance.
The Dutch team, which finished fourth in Eredivisie last year, provided better opportunities and thrilled a sell out crowd when Wittek finally proved their dominance.
Spurs' Bryan Gil was the first to cross the line after the half-time break, but they were not threatened thereafter and managed only one goal.
Nuno Espirito Sante's team is now third in Group G at the halfway point, having drawn with Rennes in France and defeated Mura, a Slovenian minnow, in their first two games.
"It was tough match against a strong team," stated Nuno, the Spurs boss. He also appointed a bench of inexperienced youngster substitutes.
"The Europa Conference League is like that, there are always difficult moments.
"We still have two home games and they are going to be a great help in solving the group.
Only group winners are automatically eligible to progress to the 16th round of Europe's third-tier club competition.
The third-placed team in the Europa League group will be playing the runners-up. The victor will then go on to the final 16.
The audition was not passed by Spurs' second string
Nuno, Spurs manager, did not consider it a risk to leave his star players back in London. He must also assess the impact of seven matches within 21 days and adjust his squad accordingly.
The Arnhem trip was the second in a busy week. His team selections showed that it was given less priority than the other games.
Spurs will face London rivals West Ham in Sunday's Premier League, and then they will head to Burnley for the fourth round of the Carabao Cup.
None of the players in the outfield who played at Newcastle made it to the Netherlands for Thursday’s match.
A number of fringe players were offered opportunities to impress their manager.
The move was a disaster as the team of nine internationals, including Harry Winks and Dele Alli from England, produced a lifeless performance.
Glenn Hoddle, Tottenham legend, questioned the attitude and stated that they should "reflect in the mirror".
The former England and Spurs manager stated on BT Sport that "they weren't up to the test".
Spurs were edgy right from the beginning as Vitesse played on Vitesse's front foot. A sell-out 24,000 crowd sparked the home side with loud and atmospheric support.
When Vittek scored the winner, the stadium burst into life. The joyous celebrations that followed - home players praising each other and soaking up the crowd's applause - were stark contrast to Spurs' defeat.
Vitesse's full-time celebrations were a sign of how important a win over an English team was to the second oldest club in the Netherlands.
