South Africa has called up Salmaan Moerat (a former SA Schools and Junior Springbok Captain).
Injury will force Faf de Klerk, World Cup winner, and Cheslinkolbe to miss South Africa's autumn Tests at Britain.
Sale Sharks scrumhalf De Klerk was injured in a hip problem and Kolbe is suffering from a knee injury.
According to some reports, De Klerk may be absent for up to five months.
As Pieter-Steph du Toit continues his rehabilitation from a shoulder injury, he was not considered for the title of world player of year.
"It is always unfortunate to lose players because of injury, but we have great depth in our squad," stated Jacques Nienaber, Springboks head coach.
South Africa will face Wales in Cardiff, Wales on 6 November and Scotland seven days later. England will follow at Twickenham on 20 Nov.
Frans Malherbe (neck), and RG Snyman(knee), are both out, while the uncapped pair Salmaan Merat and Grant Williams were called up.
De Klerk is not present, but there are four Premiership players in the squad: Marco van Staden and Jasper Wiese (Leicester back-row forwards), Vincent Koch (Saracens prop) and Lood de Jager (Sale lock).
Duane Vermuelen, a New Ulster signee, is also part of the squad.
Nienaber said: "The core team of players who played an important role in the series against British and Irish Lions as well as in the Rugby Championship will remain together as consistency and continuity are built in our squad with an eye to the 2023 Rugby World Cup."
