Raffi Quirke and Marcus Smith, fly-half and scrum-half respectively, made it through. George Ford and Billy Vunipola were also in the mix.
England's autumn internationals squad includes Raffi Quirke, uncapped Sale scrum-half, and Tommy Freeman from Northampton. Both are 20 years old.
The 34-man squad includes two more uncapped players from Gloucester, Mark Atkinson (centre) and Nic Dolly (hooker), who are both uncapped.
There is no place for Mako, Billy Vunipola and George Ford, as well as Jamie George, who are 50-cap stars.
Marcus Smith, Harlequins flyhalf, is included in the list. Owen Farrell has been confirmed to be captain.
Tonga will face England on 6 November; Australia on 13 November; and South Africa, the world champions, on 20 November.
Eddie Jones, head coach, stated that "This is a young and very talented team who have all earned our places."
"It's one of the most competitive teams I have ever selected."
Ford, who was left out of the England training squad last month, has been in great form for the unbeaten Leicester Tigers. George and the Vunipolas were at their best in Saracens’ 70-point defeat of Bath on Sunday. Sarries boss Mark McCall said afterwards that he would be "amazed” if his veteran trio were not again mentioned.
Jones insists that Jones is open to any player. However, Jones's omission of high-calibre players shows his determination to overhaul his squad, particularly given the poor Six Nations and the often-stated desire for the 2023 Rugby World Cup.
George is not there, so hooker Dolly gets a reward for his excellent early-season performances with Tigers. Ford's omission opens the door for Quins star Smith, who will be able to take the number 10 jersey.
Alex Dombrandt, Sam Simmonds, and Callum Chick can all benefit from Billy Vunipola's absence, even though Jones believes Tom Curry is still an option.
Another surprise absentee is scrum half Dan Robson. Quirke and rookie nines Harry Randall, who both won their first caps this summer, preferred him to Robson.

Quirke scored two goals against Harlequins Friday night. His boss Alex Sanderson told BBC 5 live that Quirke could become a legend in English rugby.
Sanderson stated, "If he keeps fit, he's probably going be one of our greats." "I have spoken with Eddie and he is one the smartest coaches I know. He will not only be looking out for England's best interests, but Raffi's too.
Luke Cowan-Dickie and Maro Itoje were all named, despite injuries on the weekend. Joe Launchbury, Elliot Daly, and Anthony Watson are both long-term injury absentees.
Daly is out. Max Malins and Freeman, as well as Leicester's Freddie Stewart (who made his debut in July), are all candidates for the full-back spot. Steward will likely be given an extended run in 15 shirt.
Eight players made their debuts during the summer Tests against USA and Canada. The front-row pairing of Jamie Blamire (Norway) and Trevor Davison (Norway) were joined by Chick Radwan and Randall Smith by two Falcons.
