He was 161 not out at the end of the first day of the two-test series with New Zealand in Pakistan to take his total to 2,485 runs across all formats.
The 28-year-old beat Mohammad Yousuf's previous record of 2,435.
Babar has 1,170 runs in nine Tests this year, which is more than any other player in the competition.
When Babar was on 12 and New Zealand missed the chance to dismiss him, they failed to take two chances.
