When England face Argentina on Sunday, Marcus Smith, Owen Farrell, and Tuilagi will be in the middle.
Farrell and Tuilagi are expected to be available for the game.
"Owen is doing well so far," said the backs coach.
We will know very soon that he is coming towards the end of his return to play.
Farrell missed last week's training camp in Jersey but has been working through the return to play protocols after sustaining a head injury.
While Farrell still needs to pass the final part of the process, Gleeson is happy to have his first choice in the middle.
"Marcus is a bit of a livewire and can play what he sees and you have Owen, who is very good at arranging and good at playing at the line," he said.

Tuilagi's workload is being closely managed by the England staff and he is expected to be available for the Argentina game.
Smith and Farrell are going to resume their partnership after the successful tour of Australia.

After making a rapid recovery from an elbow injury, wing Jonny May has been added to the England squad.
At least one of the three forwards will make their international debut on Sunday.
The matchday squad will be confirmed on Friday.
