She and NatSciver made 55 from 31 balls.
The tourists were dismissed for 218 in 48 overs before England never let them build any pressure during the chase.
Laura Wolvaardt made 43 before Tryon hit an 88 off 73 balls, which included three sixes, and looked as if she would propel her side to a reasonable total.
Her downfall led to a collapse of five of the team's players for just 13 runs.
In her first international appearance of the summer, Brunt took 3-18 andSciver took 4-59
The one-off Test was rained off and England lead the series 4-2.
Lamb didn't score a run in her first two matches and faced just two balls in her next match.
She showed a glimpse of her class and fine form during her knock of 38 in the Test match, and grabbed her next chance here with ease.
Lamb was able to dominate due to the lack of consistency in the South African bowling attack.
Sophia Dunkley hit back with 22 from 24 balls before she was dismissed by Nadine de Klerk.
Dunkley's departure brought inSciver, who continued her imperious form for half a century.
After centuries in the World Cup final and the Test match, Sciver took her tally to 372 runs between dismissals before she was caught by De Klerk.
The chase looked easy because Lamb andSciver scored everywhere.
There is more to come.
